چکیده

To determine the most favorable conditions for the production of ethanol by Pachysolen tannophilus, this yeast was grown in batch cultures with various initial concentrations of two of the constituents of the culture medium: o-xylose (s,), ranging from 1 g-1-r to 200 g-l-‘, and yeast extract (I,), ranging from 0 g-1-r to 8 gel-‘. The most favorable conditions proved to be initial concentrations of s, = 25 g-Z_’ and 1, = 4 g-Z_‘, which gave a maximum specific growth rate of 0.26 h I, biomass productivity of 0.023 g. I-‘. h-r, overall biomass yield of 0.094 g.g-l, specific xylose-uptake rate (qs) of 0.3 g.g-r . h-r (for t=SO h), specific ethanol-production rate (qE) of 0.065 g.g-l. h-l and overall ethanol yield of 0.34 g-g-‘; qs values decreased after the exponential growth phase while qE remained practically constant.

Mutants of Pachysolen tannophilus with Improved Production of Ethanol from d-Xylose.

The conversion of d-xylose to ethanol by the yeast Pachysolen tannophilus is relatively inefficient in batch culture. The inefficiency has been attributed in part to concurrent utilization of ethanol in the presence of appreciable concentrations of d-xylose and to the formation of xylitol and other by-products. To increase the concentration of ethanol accumulated in batch cultures, UV-induced m...

Continuous conversion of D-xylose to ethanol by immobilized pachysolen tannophilus.

The yeast Pachysolen tannophilus was entrapped in calcium alginate beads to ferment D-xylose on a continous basis in the presence of high cell densities. Experimental operating variables included the feed D-xylose concentration, the dilution rate, and the fermentor biomass concentration. Under favorable operating conditions, cultures retained at least 50% of their initial productivity after 26 ...

Characterization of Ethanol Production from Xylose and Xylitol by a Cell-Free Pachysolen tannophilus System.

Whole cells and a cell extract of Pachysolen tannophilus converted xylose to xylitol, ethanol, and CO(2). The whole-cell system converted xylitol slowly to CO(2) and little ethanol was produced, whereas the cell-free system converted xylitol quantitatively to ethanol (1.64 mol of ethanol per mol of xylitol) and CO(2). The supernatant solution from high-speed centrifugation (100,000 x g) of the ...

Enhanced Production of Xylitol from Corncob by Pachysolen tannophilus Using Response Surface Methodology

Optimization of the culture medium and process variables for xylitol production using corncob hemicellulose hydrolysate by Pachysolen tannophilus (MTTC 1077) was performed with statistical methodology based on experimental designs. The screening of nine nutrients for their influence on xylitol production was achieved using a Plackett-Burman design. Peptone, xylose, MgSO4·7H2O, and yeast extract...

Continuous ethanol production from D-xylose by Candida shehatae.

Much progress has been made since the discovery that some yeasts can ferment D-xylose to ethanol. 1 Initially, only a few yeasts were found that could produce ethanol from D-xylose. Today, at least eight species of yeasts are known to produce significant amounts of ethanol from D-xylose. 2,4-6 Three of these species have been studied extensively: Pachysolen tannophilus, Candida shehatae, and Pi...
